The alkaloids are heterocyclic compounds, generally derived from amino acids, containing one or more nitrogen atoms in its structure, which are part of the largest group of plant secondary metabolites. They are soluble both in polar and non-polar solvents, and characterized by a powerful physiological activity. On the one hand, due to their special activities, alkaloids are widely used in fields such as healthcare, cosmetics, food, and pesticides. On the other hand, some alkaloids have toxic or strong physiological effects on the living organism. Therefore, the content of alkaloids needs to be strictly monitored in various fields.

TLC is a method of separation, or identification of a mixture of components by using finely divided adsorbent solid/ liquid over a glass plate, and liquid as a mobile phase. TLC can be used to help determine the number of alkaloids in a mixture, the identity of alkaloids, and the purity of an alkaloid.
HPLC is an analytical technique used to separate, identify, or quantify each component in a mixture. HPLC is characterized by high separation efficiency, high sensitivity, low sample usage, automated online detection, low cost, and various modes. HPLC can be used in combination with UV detector, diode array detector, fluorescence detector, evaporative light detector, and electrochemical detector respectively, all of which can detect the content of alkaloids, and the results obtained are highly accurate.
GC-MS is a hyphenated analytical technique that combines the separation properties of GC with the detection feature of MS to identify different substances within a sample. GC-MS is suitable for some alkaloids that are volatile and do not decompose when exposed to heat, such as matrine, ephedrine, arecoline, etc.
UV-Vis Spectroscopy is an analytical method that can measure the analyte quantity depending on the amount of light received by the analyte. Alkaloids have certain optical activity, and their content can be determined by UV-Vis Spectroscopy.
Acid dye colorimetry mainly uses chemical reactions between alkaloids and acid dyes to produce ion pairs. The ion pairs are quantitatively dissolved in certain organic solvents, and finally the colorimetric determination can be performed. This method is generally used for the determination of total alkaloid content.
