Description | Genisteine is a natural product that is found in plants and soybeans. It has been shown to have cytostatic, antineoplastic, anti-inflammatory, and antioxidant properties. Genisteine has been shown to inhibit the growth of human skin cancer cells by binding to nitrogen atoms of guanine in DNA. The reaction mechanism appears to involve a hydrogen tartrate-mediated oxidation of genisteine followed by a nucleophilic attack on the DNA. X-ray diffraction data show that genisteine forms one or two well defined coordination geometries with the DNA molecule. In addition, structural analysis shows that genisteine binds to the phosphate group of dna polymerase chain reaction (PCR) primers and inhibits PCR activity. This inhibition may be due to steric hindrance or because genisteine competes with primer for binding sites on the template strand. Genisteine also inhibits protein synthesis by inhibiting RNA polymerase II activity in vitro and in vivo |
