· The effectiveness of nitidine as an anti-malarial agent was assessed in vitro on both chloroquine-sensitive and chloroquine-resistant strains. In vivo experiments were then conducted following the Peter's test methodology over a four-day period. To understand how nitidine operates, its impact on the parasite's cell cycle was investigated, and its localization within the parasite was determined utilizing confocal microscopy.
· Nitidine exhibited consistent in vitro efficacy against both chloroquine-sensitive and resistant strains and displayed a favorable selectivity index (> 10) when assessed against non-cancerous cell lines. Although its in vivo effectiveness was moderate, there were no indications of acute toxicity observed during the treatment. Analysis of nitidine's impact on the parasite's cell cycle revealed that it did not disrupt DNA replication, which aligned with the finding that nitidine was predominantly located in the cytoplasm rather than the nucleus of the parasite. Furthermore, nitidine formed a 1-1 complex with haem in vitro and effectively hindered the formation of β-haematin with comparable potency to chloroquine.
